This work considers the possibility of pharmacokinetic interaction between aspirin and diclofenac sodium in humans, in cases of combined intake. Possible mechanisms of such interaction are explored. It is found that aspirin causes a drop of around 75% in the Cmax and nearly 50% in the AUC of diclofenac in humans. To explain the results, two hypotheses are suggested, namely: a) that diclofenac is biotransformed pre-systemically and/or b) that diclofenac is biotransformed rapidly, to a great extent in the first pass effect, and that the presence of aspirin and/or salicylic acid can influence these processes as a catalyst.
